Description

Description: This assay employs a two-site sandwich ELISA to quantitate LGALS3 in samples. An antibody specific for LGALS3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LGALS3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LGALS3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LGALS3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Galectin-3 contains a carboxyl-terminal lectin domain and an amino-terminal non-lectin part consisting primarily of short tandem repeats. It is widely distributed in tissues and found in epithelial cells, fibroblasts, dendritic cells, and inflammatory cells.
In many cell types studied, galectin-3 is present diffusely in the cytoplasm, but is also localized to the nucleus and subcellular structures, such as mitochondria, phagosomes and exosomes, under specific conditions. It is secreted by various cell types, including monocytes, macrophages, and epithelial cells, and the extracellular protein can bind to a large number of different glycoconjugates on the cell surfaces and extracellular matrices
